Localization of CBP with Rescued and Non-Rescued Models of MJD Pathology

Abstract: Machado-Joseph disease (MJD), which is also called spinocerebellar ataxia type 3, is a progressive neurodegenerative disease. The development of the disease is associated with formation of insolulable inclusion bodies that reside within the nucleus. Additionally, it is hypothesized that MJD pathology is associated with transcription factor sequestration into nuclear inclusions thus resulting in TR dysregulation. Our studies sought to assess the effect of coexpression of human MJD with heat shock chaperones, which are known to rescue MJD pathology, on aggregate formation and localization of CBP transcription factor. Investigation of rescued and non-rescued models showed differential CBP localization within the nucleus associated with aggregate formation.
